The US Food and Drug Administration recently dealt a blow to Pfizer (PFE) and Pain Therapeutics, Inc. (PTIE) by issuing a second complete response letter (CRL) for their opioid candidate, Remoxy. Remoxy became a part of Pfizer’s pipeline following its acquisition of King Pharmaceuticals earlier this year. Remoxy has been developed, using Durect Corporation’s (DRRX) ORADUR technology, for moderate to severe pain that requires continuous, around-the-clock opioid treatment for an extended period of time. The CRL is a major setback for Pain Therapeutics and Durect Corp. While Pain Therapeutics’ shares dropped more than 40% on the news, Durect’s shares were down 31%. Second CRL for Remoxy Remoxy’s regulatory path has not been smooth with the candidate receiving a second CRL from the FDA. King Pharma had initially submitted the Remoxy

Oxecta (formerly known as Acurox) gained approval for the management of acute and chronic moderate to severe pain where the use of an opioid analgesic is appropriate. Oxecta became a part of Pfizer’s portfolio following its acquisition of King Pharma earlier this year. Oxecta has been developed under an agreement for the licensing, development, and commercialization of immediate release (IR) pain medicines utilizing Acura’s Aversion (abuse-deterrent) technology.
King Pharmaceuticals (KR) recently moved pain candidate, Eladur, which has been in-licensed from Durect Corporation (DRRX) into a phase IIb study. Eladur, a transdermal drug patch, is intended to provide continuous delivery of bupivacaine for up to 3 days from a single application, as compared to a wearing time limited to 12 hours with currently available lidocaine patches. The candidate has been developed using Durect’s proprietary TRANSDUR transdermal technology. The phase IIb 12-week, randomized, double-blind, placebo-controlled trial will study the efficacy and safety of Eladur in patients suffering from chronic low back pain. About 260 patients will participate in this study. Eladur became a part of King’s pipeline portfolio following its December 2008 acquisition of Alpharma.
Pain Therapeutics, Inc. (PTIE) recently presented encouraging data on its metastatic melanoma candidate, PTI-188. The company said that the administration of a single dose of PTI-188, a radio-labeled monoclonal antibody, resulted in the stabilization or decrease in the size of a number of melanoma tumors. In one of the phase I studies, about 70% of the patients showed stable disease in target lesions at the end of six weeks. The second phase I study showed that about 40% of the patients showed stable disease at week fourteen. PTI-188 was well-tolerated at all doses. Preliminary analysis showed that the median overall survival time based on data from both the studies was 13 months. This represents an improvement over the current median survival time of 9 months in metastatic melanoma patients. The
