Yesterday, BioMarin Pharmaceutical Inc. (BMRN) said it started a mid-stage study of PEG-PAL for the treatment of phenylketonuria (PKU). The company expects initial results from the study in mid-2010. It is co-developing the candidate with Merck Serono (MRK). The primary objective of the open-label, multi-center study, to be conducted in up to 35 patients, is to evaluate the effect of PEG-PAL on blood phenylalanine (Phe) concentrations in PKU patients. The secondary objectives are to evaluate its safety and tolerability, immune response and steady state pharmacokinetics of subcutaneous injections of multiple dose levels. BioMarin, which competes with companies such as Enzon Pharmaceuticals Inc. (ENZN), Shire Plc (SHPGY) and Altus Pharmaceuticals Inc. (ALTU), already has Kuvan in the market for treating PKU. The primary difference between PEG-PAL and Kuvan is the mode

Yesterday, Merck & Co. (MRK) announced that it is seeking approval from the European Union (EU) for its $41.1 billion acquisition of Schering-Plough Corp. (SGP). The regulatory authorities in the EU are expected to pass a verdict on the matter by Oct. 23. Merck filed for approval of the merger in the EU after the completion of the sale of its 50% interest in animal health company Merial Limited (Merial) to Sanofi-Aventis (SNY) for a cash consideration of $4 billion ( US ). As a reminder, Merial was founded in 1997. It was a 50/50 joint venture between Merck and Sanofi-Aventis and is now a wholly owned subsidiary of the latter. In 2008, Merial was ranked third in the animal health market, with a market share estimated at 13.8% and

The US Food and Drug Administration recently approved Novartis’ (NVS) Valturna for high blood pressure. The drug is a single-pill combination of Diovan (valsartan), an angiotensin receptor blocker, and Tekturna (aliskiren), the only approved direct renin inhibitor. We believe the approval is quite significant as Diovan, one of Novartis’ top-selling drugs with global sales of $5.7 billion in 2008 is losing patent protection in most European Union countries in 2011 and in the U.S. in 2012. Competition is likely to intensify shortly as Merck’s (MRK) Cozaar (2008 sales of $3.5 billion) will lose exclusivity in the U.S. in 2010, bringing in low-priced generics. The approval was based on a randomized, double-blind, placebo-controlled clinical trial on 1800 patients. The trial studied the impact of valsartan (160 mg and 320 mg) and

The European Union (EU) recently expanded the approval of Merck’s (MRK) Isentress (raltegravir) for the first line treatment of HIV patients. Earlier, it was approved for patients who had not responded to other treatment options. In the US, following a recent Food and Drug Administration (FDA) approval, Isentress can be used in combination therapy for previously untreated patients as well. Isentress recorded worldwide sales of $172 million for the second quarter of 2009, an increase of 123% compared with the year-ago period. We believe the recent FDA and EU approval (for previously untreated patients) could increase the number of targeted patients considerably, leading to a significant rise in sales. We expect the drug to record more than $1 billion in revenues by 2011. Recently, at the 49th Interscience Conference on

As a reminder, Heplisav was on clinical hold in the U.S since Mar 17, 2008 following the report of the severe adverse event (SAE) of Wegener’s granulomatosis. The SAE was reported in the phase III study in healthy adults conducted outside the U.S. Wegener’s granulomatosis is an uncommon disease which results in the inflammation of blood vessels .The vaccine is being studied for both healthy adults and patients with end-stage renal disease.
Yesterday, Amylin Pharmaceuticals, Inc. (AMLN) entered into an exclusive agreement with Indian biotech company Biocon Limited for the development, commercialization and manufacture of a novel peptide therapeutic for the potential treatment of diabetes. This agreement should ensure the optimum utilization of the synergistic capabilities of both the companies. Both companies have a strong focus on the development of treatments for diabetes. While Amylin’s “phybrid” technology will be used for the development of the candidate, Biocon will use its know-how in recombinant microbial expression to manufacture the compound. A phybrid is a peptide hybrid molecule that combines the pharmacological effects of two peptide hormones into a single molecular entity.
Yesterday, Merck (MRK) updated the status of two of its pipeline candidates, both used to treat migraine. While the company abandoned MK-3207, it will continue to develop telcagepant (MK-0974). Although MK-3207 was found to be effective, it caused liver abnormalities in some patients on discontinuing the drug. Due to this, Merck has decided to abandon any further studies of the drug. The phase III trial conducted by Merck for telcagepant found it to be more effective than placebo for acute treatment of multiple migraine attacks. In another study, the safety of the product was compared to another migraine drug of Merck, Maxalt (rizatriptan). The company presented data from both trials at the International Headache Congress and plans to initiate discussions regarding the next steps with the US Food and Drug

Originally, Gardasil was approved by the FDA in June 2006, as the first vaccine for cervical cancer, caused by human papillomavirus (HPV) for females 9 to 26 years of age. The panel voted 7-1 in favor of the vaccine’s safety and 7-0 (with one abstention) regarding its effectiveness. Although the FDA is not required to follow the panel’s recommendation, it generally does so. The final decision from the agency is expected by year end. While the advisory committee voted 12-1 in favor of the efficacy of the vaccine, its safety profile got 11-1 in support from the panel. While the FDA is not bound by the advisory committee’s recommendation, it generally follows the suggestion. Cervarix was found to be highly effective and was well tolerated in the prevention of cervical pre-cancers and cervical cancer related to human papillomavirus (HPV) types 16 and 18, in girls and young women. While the fixed-dose combination of mometasone furoate and formoterol fumarate for the maintenance treatment of asthma in patients 12 years and above is currently under review by the U.S. Food and Drug Administration (FDA), the filing for FDA approval of the contraceptive is expected in 2010. The oral contraceptive is a combination of a natural estrogen (which is identical to the estrogen produced by a woman’s body) and a selective progestin. The drug was licensed from Merck KGaA. The company is seeking EMEA approval for the drug to address the unmet need for patients suffering from acute atrial fibrillation (AF, abnormal heart rhythms leading to strokes). The deal, which is expected to close by the fourth quarter of this year, has been approved by both company boards. Additionally, Neurogen shareholders, representing approximately 33% of outstanding shares, have already given their nod in favor of the deal.
Yesterday, the U.S. Food and Drug Administration (FDA) approved GlaxoSmithKline’s (GSK) Hiberix, a Haemophilus influenzae Type b (Hib) vaccine. The vaccine has been approved as a booster dose for children in the age group of 15 months to 4 years. FDA approval was based on safety and efficacy data obtained from 7 clinical studies conducted in Europe, Latin America and Canada involving more than 1,000 children. Hiberix was approved under the FDA’s accelerated approval pathway, mainly due to a nationwide shortage of Hib vaccines since December 2007. Merck & Co., Inc.
Merck (MRK) received a favorable ruling in a major patent-infringement suit yesterday against the largest generic player, Teva Pharmaceuticals (TEVA), on its top-selling drug Singulair. Teva was seeking the Food and Drug Administration’s (FDA) approval for selling a generic version of the drug for asthma and allergic rhinitis. The litigation has been continuing since Merck sued Teva in February 2007. During the second quarter of 2009, Singulair recorded worldwide sales of $1.3 billion, up 16% from the year-ago period. The drug had total sales of $4.3 billion in fiscal 2008, accounting for about 18% of Merck’s total revenue. Defending Singulair’s patent was thus quite crucial for Merck. We believe this is a major win for Merck as an unfavorable ruling would have hurt its top-line badly. The court not only

The ministry accepted the Mashelkar committee’s report, according to which India will continue to allow patents on incremental innovations if the enhancements are beneficial to patients. The decision was taken amidst severe protests from generic players and health activists who fear this will not allow cheaper generic drugs to enter the market. On the basis of 2008 sales figures, the Indian pharmaceutical market ranks 13th in the world, with sales of $7.7bn last year (an increase of 4% over 2007).
Yesterday, the U.S. Food and Drug Administration’s (FDA) advisory panel ruled in favor of Amgen’s (AMGN) osteoporosis drug denosumab (proposed trade name: Prolia), but with certain exceptions. While the advisory panel unanimously gave its stamp of approval to the injectable drug for prevention of bone fractures in women with postmenopausal osteoporosis, it stated that the drug should only be used by patients facing the highest risk of fractures. The ruling was based on safety and efficacy data from 30 studies of the drug involving more than 12,000 patients. However, the panel did not approve use of the drug in the prevention and treatment of bone loss in women with breast cancer undergoing hormone ablation. It demanded additional data to ensure that denosumab would not hasten tumor growth in such patients,

Yesterday, Schering-Plough Corp. (SGP) said it has signed an agreement with Orchid Chemicals & Pharmaceuticals Ltd. and Orgenus Pharma Inc., which sought to make generic versions of its allergy drug Clarinex. This signals the end of all pending litigations that Schering filed against several generic drug manufacturing companies in the US District Court of New Jersey since 2006. These companies wanted to market solid oral doses of the drug, desloratadine, in the U.S. as generic brands before the expiration of certain Schering patents. Pursuant to the agreement, Orchid received rights to launch a generic version of the Clarinex brand Reditabs in the U.S. on Jan. 1, 2012 and Clarinex brand 5 milligram tablets on July 1 of that year. Schering holds various patents related to Clarinex till 2022. Schering has

GlaxoSmithKline (GSK) recently entered an agreement with Nabi Biopharmaceuticals (NABI) to bring a new pipeline candidate into its vaccine portfolio. Glaxo will buy Nabi’s PentaStaph vaccine, which is being developed to fight hospital-acquired staph infection. Staph infections are caused by a drug-resistant strain of bacteria often found in hospitals. PentaStaph is being tested against three different strains of staph and two other antigens. The deal is valued at $46 million, of which $20 million will be paid immediately. The remaining $26 million will be paid after achievement of four key milestones. The deal is scheduled to close by year-end and Nabi expects to achieve the four milestone payments within 18 months. GlaxoSmithKline has a strong presence in the vaccine market.
by The Investment U Research Team The mood seems to have soured on Wall Street since its bounce early this morning. And if you peer beneath the headlines, you can see why. The big news has been Caterpillar’s (NYSE: CAT) earnings, which tripled analyst’s expectations. But as we look deeper, we find that CAT’s earnings are well below last year’s figures. Merck (NYSE: MRK) announced that earnings dropped less than expected. Again, note revenue still dropped 2%. State Street (NYSE: STT) reported a loss of almost $3.3 billion as it writes down asset values. Coca-Cola (NYSE: KO) also announced lower earnings, citing stronger dollar. On the surface, these earnings reports sound good, but they hide the ugly truth. And the list goes on, from BlackRock (NYSE: BLK), Freeport-McMoRan (NYSE: FCX)

by The Investment U Research Team A couple of interesting news items came out in the past few days. One, a research report showing how expensive treatment is for childhood mental issues. It also showed that in 2006 over 13 million children were treated for asthma at a cost of $8 billion. The other a study that finds more Americans than ever are taking some sort of prescription for mental illness – 73% more than in 1996. What’s interesting, apart from the fact that we seem to be overmedicating as a society, is that the numbers of patients and the costs of treatments spell hundreds of billions in profits for healthcare and drug makers. Taking a look at the eleven largest, we find that their market caps total almost $900

OVERVIEW The financial market and the economic landscape in the U.S. have not changed much since the beginning of the year. And we believe the status quo will last for a while at least in 2009. Large biotech companies are doing OK since they have strong balance sheets to weather the current market turmoil and can generate revenue from product sales. Investors in large biotechnology companies worth more than $1 billion have preserved much of their wealth. Smaller biotech companies, particularly those still in development stage, meanwhile, are under severe strain as the global economic slump cuts into sales and their ability to raise fresh cash to fund research. As we expected, the M&A spree has continued in the first three months of this year. Following the acquisition of Wyeth
