(PLX) was dealt a huge blow recently with the US Food and Drug Administration (FDA) issuing a complete response letter (CRL) for the company’s Gaucher disease candidate, taliglucerase alfa. The next five years are expected to reflect a significant imbalance between new product introductions and patent losses. According to IMS Health, this is the main reason global pharmaceutical market growth will be restricted to the mid-single digits (5-8%) through 2014. Over the next five years, products that currently generate more than $142 billion in sales are expected to face generic competition, including Lipitor, Plavix and Zyprexa. In fact, 2011 itself will see products worth more than $30 billion losing patent protection. This includes products like Lipitor, Plavix, Zyprexa and Levaquin.
With 2010 coming to an end, the pharmaceutical industry continues to witness major challenges like sluggish prescription trends, EU pricing pressure, intensifying generic competition and limited late-stage catalysts. The next five years are expected to reflect a significant imbalance between new product introductions and patent losses. According to IMS Health, this is the main reason global pharmaceutical market growth will be restricted to the mid-single digits (5-8%) through 2014. Over the next five years, products that currently generate more than $142 billion in sales are expected to face generic competition Incorporatedluding Lipitor, Plavix and Zyprexa. In fact, 2011 itself will see products worth more than $30 billion losing patent protection. This includes products like Lipitor, Plavix, Zyprexa and Levaquin. According to IMS Health, these products generated more than $17 billion

French pharmaceutical company, Sanofi-Aventis (SNY) recently presented encouraging results on its multiple sclerosis (MS) candidate, teriflunomide. Results from a phase III randomized, double-blind, placebo-controlled study (TEMSO) showed that teriflunomide helped reduce annualized relapse rates by 31% compared to placebo. The study evaluated two doses of teriflunomide – 7 mg and 14 mg. Sanofi said that treatment with teriflunomide helped reduce the risk of disability progression by 30% and 24% for the 14 mg and 7 mg doses, respectively. The candidate was found to be well-tolerated. Sanofi also presented long-term safety data on relapsing multiple sclerosis patients. Eight years follow-up data from a phase II study showed that the candidate was well-tolerated. The successful development of teriflunomide could provide multiple sclerosis patients with a new first-line treatment option. Teriflunomide is currently

French pharma giant Sanofi-Aventis (SNY) recently went hostile with its bid to acquire biotech company Genzyme Corporation (GENZ). Sanofi has launched a tender offer for all outstanding shares of Genzyme – the offer is scheduled to expire on Dec 10. Sanofi’s offer price stands at $69 per share with the entire deal being valued at $18.5 billion. The $69 offer price represents a 38% premium over Genzyme’s share price on July 1, 2010. Rumors regarding Sanofi’s intention to acquire Genzyme have been doing the rounds since early July 2010. Since then, Genzyme’s shares have shot up more than 40%. Sanofi said that it launched the hostile bid mainly due to Genzyme management’s refusal to enter into constructive discussions with Sanofi.
We are downgrading Cell Therapeutics, Inc. (CTIC) to Underperform from Neutral with a price target of $0.25 primarily due to the uncertainty surrounding the US approval of its lead candidate, pixantrone. Pixantrone (proposed trade name: Pixuvri), is currently under development for the treatment of relapsed or refractory aggressive non-Hodgkin’s lymphoma (NHL) in patients who have not responded to other treatment options. Cell Therapeutics Inc., based in Seattle, Washington, is focused on developing drugs for cancer. The company received a setback in April 2010, when the US Food and Drug Administration (FDA) denied approval to pixantrone and issued a complete response letter (CRL) based on issues related to the study design. The FDA asked Cell Therapeutics to conduct an additional trial to determine the safety and efficacy of pixantrone. In August

The decision to approve the product does not come as a surprise as the drug’s approval for the long-term treatment of type I Gaucher disease was recommended by the European Medicines Agency’s (EMA) Committee for Medicinal Products for Human Use (CHMP) in June 2010. Vpriv, an enzyme replacement therapy, enjoys orphan drug status from the Committee for Orphan Medical Products. Vpriv is already available in the US where it received approval from the US Food and Drug Administration in March 2010. The committee recommended the approval of the enzyme replacement therapy for treating type I Gaucher disease. Furthermore, the drug has been granted accelerated assessment by the EMA in the European Union (EU) and enjoys orphan drug status from the Committee for Orphan Medical Products. Shire should be in a position to launch Vpriv in the EU by year end. Meanwhile, product launch in other countries should commence in 2011.
Following the release of first quarter results on April 21, 2010, several analysts covering Genzyme Corporation (GENZ) have lowered their earnings estimates for 2010 and 2011. Genzyme is still suffering from the impact of the temporary shutdown of its Allston Landing manufacturing facility last year and has yet to resume normal supply of the key products that were affected by the shutdown. First Quarter Highlights Genzyme reported first quarter earnings of 24 cents per share, above the Zacks Consensus Estimate of 19 cents but well below the year-ago earnings of 70 cents. With the company facing manufacturing problems from mid-2009, we were not surprised to see first quarter revenues decline 6.5% to $1.07 billion, mainly due to restricted supplies of Cerezyme and Fabrazyme.
Isis Pharmaceuticals Inc. (ISIS) reported a net loss of 10 cents per share in the first quarter, well below the Zacks Consensus Estimate of a net loss of 17 cents. However, the reported quarter’s loss was higher than the year-ago period’s net loss of a cent mainly due to lower revenues and higher operating expenses. Following the adoption of a new required accounting standard, Isis is no longer including Regulus Therapeutics’ financial results. Regulus has been established in collaboration with Alnylam Pharmaceuticals (ALNY). First quarter revenues, which include license fees, milestone-related payments and other payments, declined 5.2% to $29.9 million.
Genzyme (GENZ) has been under a lot of pressure over the past few quarters following the temporary shutdown of its Allston manufacturing facility due to contamination problems. While we are pleased to see that Genzyme is taking steps to emerge from the impact of the temporary shutdown of the Allston plant, we believe that the company may face additional challenges before it is able to go back to a normal production and supply schedule. Moreover, the FDA is looking to enforce a consent decree on the company which would result in Genzyme incurring additional costs. With the company yet to emerge fully from its manufacturing issues, we are downgrading the stock to Underperform. Although Genzyme could receive some positive news in the form of FDA approval of Lumizyme, we expect

Isis Pharmaceuticals, Inc. (ISIS) and Genzyme Corp. (GENZ) recently announced the publication of phase III data on mipomersen in The Lancet. The data was published from a study that was conducted in patients with homozygous familial hypercholesterolemia (hoFH). Results, which were presented at the annual American Heart Association meeting in November 2009, showed that mipomersen met its primary endpoint. The candidate achieved a 25% reduction in low-density lipoprotein – cholesterol (LDL-C) in an intent-to-treat analysis. Besides meeting the primary endpoint, the study also met each of its secondary and tertiary endpoints, which included statistically significant reductions in apolipoprotein-B, total cholesterol, non- high-density lipoprotein (HDL) cholesterol, lipoprotein (a) [Lp(a)], very low density lipoprotein-cholesterol (VLDL-C) and triglycerides. Mipomersen is Isis’ lead pipeline candidate. It is an antisense drug that targets apolipoprotein B

The company had reported a net loss of 8 cents in the year ago period. The wider-than-expected loss was due to higher operating expenses. For the full year, the company reported a net loss of 31 cents, well above the net loss of 10 cents reported in 2008. Once again, higher operating expenses accounted for the wider loss despite an increase in revenues. Fourth quarter revenues increased 8.8% to $32.3 million. Isis and partner, Genzyme Corp. (GENZ) recently announced positive results from a phase III study. Data showed that patients suffering from heterozygous familial hypercholesterolemia (heFH) experienced a 28% reduction in low density lipoprotein (LDL) or “bad” cholesterol after 26 weeks of treatment with mipomersen. In contrast, patients receiving placebo experienced a 5% increase in cholesterol levels. Although these results were encouraging and statistically significant, concerns regarding the candidate’s safety profile surfaced with the company reporting elevations in liver transaminases, which is a sign of possible liver damage. While the company stated that the elevations were similar to that observed in earlier studies, we remain concerned that concerns regarding

The pharmaceutical industry has witnessed major changes over the past few quarters, with performance being affected by factors like sluggish prescription trends, intensifying generic competition and limited phase III catalysts. The next five years are expected to reflect a significant imbalance between new product introductions and patent losses. According to IMS Health (RX), this is the main reason global pharmaceutical market growth will be restricted to the mid-single digits through 2013. Over the next five years, products that currently generate about $137 billion in sales are expected to face generic competition Incorporatedluding Lipitor, Plavix and Seretide. At the same time, new products are not expected to generate the same level of sales as products losing patent protection. With revenue growth stalling or slowing down, companies have been resorting to cost-cutting

Genzyme is seeking approval for Lumizyme which has been produced at its 4000 liter-scale manufacturing plant in Geel, Belgium. Earlier, Genzyme had submitted an application seeking approval for Lumizyme manufactured at its 2000 liter bioreactor scale at its Allston Landing manufacturing facility. However, the FDA issued a complete response letter in November 2009 stating it will not grant approval unless the company addresses the manufacturing issues at its Allston plant.
Genzyme Corp. (GENZ) reports fourth-quarter results on February 17, with the current Zacks Consensus Estimate at 20 cents per share. Recent revisions to the quarterly estimate have been in both directions, with a very modest upward (positive) bias. Of the 12 estimates for the fourth quarter, three have been raised while two have been lowered in the last 30 days. On balance, the estimate is up from 18 cents per share in the last 30 days. If quarterly results come inline with expectations, as is most likely given its recent track record, fourth-quarter 2009 EPS would be 52.4% below the year-earlier level. The year-over-year drop is due to the lingering effects of operational issues at the company’s Allston Landing, Massachusetts facility. For the full-year 2009, the current Zacks Consensus Estimate

But that’s the overall vibe that I took away from the JP Morgan Healthcare Conference. The investors I spoke with reminded me of jilted lovers – eager to get back on the market, but scared to get hurt again. Rather than checking out the eligible candidates on Match.com, though, healthcare investors spend their time poring over companies’ balance sheets and drug pipelines. And so it was in San Francisco.
