The pharmaceutical industry continues to be impacted by major issues such as sluggish prescription trends, European Union (EU) pricing pressure, intensifying generic competition and limited late-stage catalysts. The next five years are expected to witness a significant imbalance between new product launches and patent expirations. All these factors will lead to a slowdown in global pharmaceutical market growth in the next five years. Major revenue generating drugs like Lipitor, Plavix, Lexapro and Zyprexa are expected to confront generic competition over the next five years. In fact, revenues in excess of $30 billion are expected to be lost in 2011 itself due to key drugs like Lipitor and Zyprexa going off-patent. The impact of the genericization of these drugs will be felt significantly in 2012, which will be a challenging year

(MYL) recently announced that Endo Pharmaceuticals (ENDP) filed a patent infringement lawsuit in connection with an abbreviated new drug application (ANDA) filed by Mylan with the US Food and Drug Administration (FDA) for the generic version of Endo’s acute migraine drug Frova. Mylan believes it is the first company to file an ANDA for generic Frova and is expecting to be granted 180 days of marketing exclusivity once approved. Frova generated revenues of approximately $68.2 million in the trailing twelve months ending on June 30, 2011. This month, Mylan launched the generic version of Uroxatral tablets. Some important products launched by Mylan in the recent past include generic versions of AstraZeneca‘s (AZN) Entocort, Novartis‘ (NVS) Femara and Cephalon‘s (CEPH) Amrix. Top line results from the study revealed that Endo Pharma’s candidate failed to meet the predetermined end points. The randomized, double blind, two-arm, placebo-controlled study evaluated the candidate in patients suffering from moderate-to-severe chronic low back pain. The Dow inched closer to the levels of February 18, which was the highest close since June 5, 2008. The Dow Jones Industrial Average (DJIA) is en route to its best quarter performance and is within striking distance of reaching its highest levels for 2011.
We are downgrading Cell Therapeutics, Inc. (CTIC) to Underperform from Neutral with a price target of $0.25 primarily due to the uncertainty surrounding the US approval of its lead candidate, pixantrone. Pixantrone (proposed trade name: Pixuvri), is currently under development for the treatment of relapsed or refractory aggressive non-Hodgkin’s lymphoma (NHL) in patients who have not responded to other treatment options. Cell Therapeutics Inc., based in Seattle, Washington, is focused on developing drugs for cancer. The company received a setback in April 2010, when the US Food and Drug Administration (FDA) denied approval to pixantrone and issued a complete response letter (CRL) based on issues related to the study design. The FDA asked Cell Therapeutics to conduct an additional trial to determine the safety and efficacy of pixantrone. In August

Cephalon, Inc. (CEPH) recently announced the completion of its acquisition of biotechnology company Ception Therapeutics, Inc. With this acquisition, Cephalon will gain access to Ception’s lead product, Cinquil (reslizumab), an investigational humanized monoclonal antibody (mAb) against interleukin-5 (IL-5). Earlier this year, Ception reported positive results on Cinquil from a phase II study that was conducted in adults with eosinophilic asthma. Results showed that Cinquil improved asthma control in adult patients with moderate to severe asthma and eosinophilic airway inflammation. Moreover, Cinquil also achieved statistical significance compared to placebo in an analysis of FEV1, which is a measure of lung function. Based on these results, Cephalon decided to exercise its option to acquire Ception.
The U.S. Food and Drug Administration (FDA) recently informed Cephalon, Inc. (CEPH) that it will require more time to review the supplemental new drug application (sNDA) submitted by the company for Nuvigil. Cephalon had filed the sNDA in June 2009, seeking to expand Nuvigil’s label as a treatment for improving wakefulness in patients with excessive sleepiness associated with jet lag disorder due to eastbound travel. The FDA had initially granted priority review status to the candidate which means a decision would have been out by Dec 29, 2009. However, with the agency asking for additional review time, a decision is now expected by March 29, 2010. Nuvigil (armodafinil) is a follow-on version of Provigil (modafinil) which contributed about 50% to revenues in 2008. Cephalon received FDA approval for Nuvigil in

by Sheena Martin, Contributing Editor Wednesday, December 16, 2009 One company looking forward to the end of 2009 is Cephalon (Nasdaq: CEPH). And that’s an understatement. The biopharmaceutical company’s share price has tumbled by 23% this year, following a slide in sales. On top of that, clinical results for a drug that treats inflation of the esophagus in children were disappointing. And generic drugs threaten its other flagship products. However, the growth potential is still significant, given promising trial results for its new cancer drug… Cephalon’s $1 Billion Drug Blockbuster In March 2008, Cephalon’s cancer treatment drug, Treanda, showed good results in patients with chronic lymphocytic leukemia – a cancer of the soft tissue inside bones where blood cells are made.
Cephalon, Inc. (CEPH) oncology product Treanda could see a significant rise in demand thanks to encouraging data presented at the recently conducted annual meeting of the American Society of Hematology (ASH). Clinical data supporting the use of Treanda as a new standard of care in the first-line treatment of non-Hodgkin’s lymphoma (NHL) was presented at ASH. Data from an investigator-led proof-of-concept study demonstrated that newly diagnosed NHL patients who were treated with a combination of Treanda and Roche/Genentech‘s Rituxan experienced superior progression-free survival (55 months) compared to patients treated with the current standard of care, Rituxan plus a chemotherapy cocktail known as CHOP (35 months). The Rituxan-Treanda therapy was also found to be safer, less toxic and better tolerated. Treanda is currently approved for the treatment of patients suffering from

Genzyme Corp. (GENZ) recently presented encouraging interim results on its oncology product, Campath, at the annual meeting of the American Society of Hematology (ASH). Results from the randomized phase III study (CAM314) showed that Campath, when used in combination with Fludara (FluCAM), significantly reduced the risk of disease progression or death compared to single-agent Fludara in patients with relapsed and refractory chronic lymphocytic leukemia (CLL). Interim results showed that the FluCAM arm demonstrated superior progression free survival (PFS) compared to Fludara alone (29.6 months versus 20.7 months), reducing the risk of disease progression or death by 39%. Importantly, in a pre-specified subgroup of patients with advanced CLL, the median PFS was more than doubled with FluCAM compared to Fludara alone (26.1 months versus 12.1 months). The FluCAM regimen provided significantly

Cephalon, Inc. (CEPH) recently reported disappointing results on a pipeline candidate, Cinquil that was being developed for the treatment of pediatric eosinophilic esophagitis (EoE). Results from the phase IIb/III study showed that Cinquil achieved a statistically significant reduction in esophageal eosinophil levels versus placebo, which was one of the co-primary endpoints. However, although patients on Cinquil showed an improvement in clinical symptoms, which was the second co-primary endpoint, the candidate failed to show a statistically significant improvement for this endpoint. Eosinophilic esophagitis is a rare disease that is more frequently diagnosed in children. The Journal of Clinical Investigation estimates that approximately 80,000 children in the United States have EoE. Cinquil is being developed by Ception Therapeutics, Inc., a privately held biopharmaceutical company and Cephalon holds an option to acquire Ception.

Barr was looking to market a generic version of Cephalon’s cancer pain management drug, Fentora. According to Cephalon, Fentora’s patent is not set to expire before 2019.
