We recently reiterated our Neutral recommendation on Biogen Idec (BIIB). Biogen is the market leader in therapies for the treatment of multiple sclerosis (MS). We believe the company will continue to retain a leading position in the MS market. Biogen’s largest product, Avonex, posted sales of $2,518.4 million, up 8.4%, in 2010. We expect Avonex to continue contributing significantly to the top-line thanks to price increases, focused marketing effort, expansion into new markets and positive clinical data. Meanwhile, Tysabri, the company’s second MS product, continues to see an increase in patients despite carrying a “black box” warning for the risk of progressive multifocal leukoencephalopathy (PML) and other cautionary language. Tysabri posted $1.2 billion in sales in 2010. Tysabri’s launch in new markets like India, Brazil, Argentina, the Middle East and

Alnylam Pharmaceuticals Inc (ALNY) recently announced pre-clinical data from a trial evaluating its pipeline candidate ALN-HTT for the treatment of Huntington’s disease (HD). ALN-HTT is a novel therapeutic product that uses the potentially revolutionary RNA (ribo nucleic acid) interference (RNAi) technology. The data showed broad central nervous system (CNS) distribution and robust therapeutic silencing of the gene responsible for Huntington’s disease. Further, ALN-HTT was well tolerated in these studies. ALN-HTT is being developed in collaboration with Medtronic, Inc. (MDT) and the CHDI Foundation. Alnylam expects to file an investigational new drug application (IND) application for the candidate in 2012. Approval of the IND by the US Food & Drug Administration (FDA) will allow the candidate to enter early stage clinical trials. Besides ALN-HTT, Alnylam is developing ALN-RSV01 for the treatment

With 2011 coming to an end, the pharmaceutical industry continues to face challenges like sluggish prescription trends, EU pricing pressure, intensifying generic competition, pipeline failures and limited late-stage catalysts. The next five years are expected to reflect a significant imbalance between new product introductions and patent losses. All these factors will lead to a slowdown in global pharmaceutical market growth in the next five years, with major revenue-generating drugs like Lipitor, Plavix, Lexapro and Zyprexa losing exclusivity. In fact, by the end of 2011, drugs worth more than a total of $30 billion will lose patent protection. This includes drugs like Lipitor, Zyprexa and Levaquin. The effect of the genericization of these products will be felt mostly in 2012, which will be a challenging year for several companies. At the

Alnylam Pharmaceuticals Inc. (ALNY) recently presented data from an early stage study of its pipeline candidate ALN-TTR01 at a scientific conference in Japan. ALN-TTR01 is a systemically delivered RNA interference technology (RNAi) therapeutic that employs a first-generation lipid nanoparticles (LNP) formulation to treat transthyretin (TTR) mediated amyloidosis (ATTR). Data from the trials showed that ALN-TTR01 led to statistically significant reductions in serum TTR protein levels for the treatment of ATTR. Overall, the candidate has proved to be well tolerated with no serious adverse events observed in the study. We believe the encouraging data further highlights the potential of RNAi as a new class of therapeutics. This is the first time an RNAi has silenced a disease-causing gene in clinical trials. ALN-TTR01 is part of Alnylam’s core product strategy, Alnylam 5×15,

by Marc Lichtenfeld, Investment U Senior Analyst Wednesday, August 17, 2011: Issue #1580 The market’s violent sell-off over the past two weeks took down many good stocks along with the bad ones. Even with the quick bounce, quality companies are still trading at steep discounts to where they were just two weeks ago and are now at very attractive valuations. This is especially true in the healthcare sector, where names like Gilead Sciences (Nasdaq: GILD), which is down 12 percent, are still below where they were trading prior to the debt crisis decline. Whether the United States’ debt is rated AAA or AA+ should have no bearing on sales of Gilead’s leading HIV drugs or whether its newest drug, Quad, will receive FDA approval. The stock trades at just 11.2 times

Merck KGaA (MKGAF) recently announced its plans to withdraw the regulatory applications filed with the US Food and Drug Administration (FDA) and the European Medicines Agency (EMA), seeking approval to market cladribine as a treatment for relapsing-remitting multiple sclerosis (MS). This decision comes after Merck KGaA’s discussions with the FDA at the end-of-review meeting related to the new drug application (NDA) for cladribine. Merck KGaA believes that data from the ongoing clinical trials will not be sufficient to address the FDA’s requirements for approval. The company is of the view that to gain US and EU approval, it will have to conduct a new clinical trial program that would take several years to complete. Cladribine is currently marketed as Movectro in Australia and Russia for the treatment of relapsing-remitting MS.

Acorda Therapeutics, Inc. (ACOR) recently reported an analysis of data on Ampyra (dalfampridine) at the Consortium of Multiple Sclerosis Centers (CMSC) annual meeting. The analysis demonstrated improvements in walking ability of patients on Ampyra as measured by patient self-report on the 12-Item Multiple Sclerosis Walking Scale (MSWS-12). The analysis comprised data from a phase II trial and two phase III trials of Ampyra. These data were a part of Acorda’s new drug application (NDA) submission to the US Food and Drug Administration (FDA) for the approval of the drug.
Challenges like sluggish prescription trends, EU pricing pressure, intensifying generic competition, pipeline failures and limited late-stage catalysts continue to impact the pharmaceutical industry. The next five years are expected to reflect a significant imbalance between new product introductions and patent losses. All these factors will lead to a slowdown in global pharmaceutical market growth in the next five years. Products like Lipitor, Plavix, Lexapro and Zyprexa are expected to face generic competition over the next five years.
French pharmaceutical company, Sanofi-Aventis (SNY) recently presented encouraging results on its multiple sclerosis (MS) candidate, teriflunomide. Results from a phase III randomized, double-blind, placebo-controlled study (TEMSO) showed that teriflunomide helped reduce annualized relapse rates by 31% compared to placebo. The study evaluated two doses of teriflunomide – 7 mg and 14 mg. Sanofi said that treatment with teriflunomide helped reduce the risk of disability progression by 30% and 24% for the 14 mg and 7 mg doses, respectively. The candidate was found to be well-tolerated. Sanofi also presented long-term safety data on relapsing multiple sclerosis patients. Eight years follow-up data from a phase II study showed that the candidate was well-tolerated. The successful development of teriflunomide could provide multiple sclerosis patients with a new first-line treatment option. Teriflunomide is currently

We are downgrading Cell Therapeutics, Inc. (CTIC) to Underperform from Neutral with a price target of $0.25 primarily due to the uncertainty surrounding the US approval of its lead candidate, pixantrone. Pixantrone (proposed trade name: Pixuvri), is currently under development for the treatment of relapsed or refractory aggressive non-Hodgkin’s lymphoma (NHL) in patients who have not responded to other treatment options. Cell Therapeutics Inc., based in Seattle, Washington, is focused on developing drugs for cancer. The company received a setback in April 2010, when the US Food and Drug Administration (FDA) denied approval to pixantrone and issued a complete response letter (CRL) based on issues related to the study design. The FDA asked Cell Therapeutics to conduct an additional trial to determine the safety and efficacy of pixantrone. In August

We recently downgraded Biogen Idec, Inc. (BIIB) to Neutral from our previous Outperform recommendation. Biogen is one of the world’s largest biotechnology companies. Its main areas of focus include neurology, oncology, hemophilia and immunology. Products such as Avonex (for multiple sclerosis), Rituxan (for non-Hodgkin’s lymphoma), and Tysabri (for multiple sclerosis) currently drive the top line. Biogen is the market leader in therapies for the treatment of multiple sclerosis (MS). We believe the company will continue to retain a leading position in the MS market. Biogen’s largest product, Avonex, posted sales of $2,322.9 million, up 5.5%, in 2009. We expect Avonex to continue contributing significantly to the top-line thanks to price increases, focused marketing effort and positive clinical data. Meanwhile, Tysabri, the company’s second MS product, continued to see an increase

Novartis AG (NVS) received a huge boost recently with the US Food and Drug Administration (FDA) approving its oral multiple sclerosis (MS) drug, Gilenya. Gilenya is the first oral treatment to gain US approval for the first-line treatment of relapsing forms of MS. The drug is shown to significantly reduce relapse rates and delay disability progression. The FDA approved Novartis’ drug along with a Risk Evaluation and Mitigation Strategy (REMS) which will inform patients and physicians on the safe use and risks associated with the drug. The FDA has recommended that patients should be monitored for a decrease in heart rate on starting treatment with Gilenya. The drug has also been seen to increase the risk of infections. The agency also recommended ophthalmologic evaluation as instances of serious eye problems

Revenue increased 11% to $1.2 billion, with Tysabri and Rituxan being the primary growth drivers. Based on second quarter results and Biogen’s new guidance for 2010, we have raised our earnings estimate for 2010 by 29 cents to $4.73. Key products Avonex and Tysabri continue to contribute significantly to sales and we expect Biogen to maintain its leading position in the multiple sclerosis market. The company is working on building its pipeline through acquisitions and in-licensing deals. In our opinion, Biogen has the best pipeline in all of biotech, and could be an attractive takeover candidate for pharma companies interested in biologics.
